Pretty similar to how you would validate it for blood. Do you have another lab that you could use for comparison who has already validated it? Alternatively, you could count cells manually and compare against the automated count.
There are also some notable CAP checklist items that pertain to body fluids that inspectors tend to give scrutiny. Example of a validation study:
