amino acids: must know one-letter symbols?

Do we need to the 1-letter abbreviations or is knowing the 3-letter abbreviations good enough?

Neither. If you're trying to memorize the individual abbreviations for each amino acid, you're either A. focusing on the wrong information, or B. you already know all the broader higher yield concepts in medicine and you should have taken the test already.
In other words, stop focusing on the minutia. It'll be more helpful if you remember that sickle cell is due to the substitution of glutamate by valine.
